Introduction

Matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometer is a fundamental tool for the identification and analysis of peptides and proteins. MALDI-TOF MS is used to identify proteins by matching a list of experimental peptide masses with the calculated list of all peptide masses of each entry in a database. The sequencing capability of MALDI-TOF/TOF makes protein identifications more specific than those obtained by simple peptide-mass mapping.

The Mass Spectrometry Facility in IMB now offers service for IMB members on sample spotting onto target and peptide or protein analysis. The facility also provides computer termini for peptide mass fingerprinting (PMF) and/or MS/MS ions searching against database.

輔助雷射脫附游離飛行式質譜儀為分析及辨識蛋白質和胜肽的主要儀器之一。例如,未知蛋白質經蛋白質酵素裂解後,藉質譜儀可得到裂解產生之各種胜肽的分子量。它們可和資料庫中已知蛋白質預測之胜肽的分子量比對,比對之結果可用以辨識該未知蛋白質。此外,利用酵素裂解的胜肽進行MALDI-TOF/TOF的定序分析,所得到的胜肽序列可以更進一步証實蛋白質辨識的正確性。

質譜儀室目前提供分生所人員樣品點樣及上機分析的服務。同時也提供電腦及資料庫供使用者做資料搜尋之用。
