Gene Targeting

Gene targeting experiment is designed to disrupt or modify an endogenous gene.  Gene targeting experiment is performed in mouse embryonic stem (ES) cells. With appropriate design, a targeting vector consists of two homologous fragments which are homologous to the gene of interest is delivered into the ES cells. With the homologous recombination events taking place and suitable drugs selections, the drug-resistant ES clones can be isolated individually. Successfully gene-targeted ES clones can be identified using genomic DNA analysis approached, i.e. PCR or Southern Blot.
