Laser Scanning Confocal Microscope

雷射掃描共軛焦顯微鏡技術(Laser Scanning Confocal Microscope)是以特定波長的雷射做為激發光源,激發樣品中的螢光分子,經由針孔(pinhole)濾波的原理去除非焦平面的訊號,保留光切層的訊號,而切層或焦平面的厚度則由Pinhole開口大小來決定,即二維影像(2Dimage)。可以搭配在正立顯微鏡、倒立顯微鏡或是電生理型的顯微鏡,進行不同的實驗設計。此類顯微鏡的應用基本上涵蓋所有的螢光觀察,對於較厚的組織與樣品,進行Z堆疊後,可以重建立體的影像,也可以在細胞培養箱中記錄長時間的螢光變化觀察。
